SDT is a developer and manufacturer of high performance reagents for ImmunoAssays, Nucleic Acid Hybridization and ImmunoHistoChemistry. Since beginning of 90s we are bulk OEM supplier of specialty products to IVD Industry and customers in Pharmaceutical Drug Discovery and HTS Programs. Of course, we also have been serving Life Science Research Community. Our specialty is production of reagents for Ultra-Sensitive Detection in ELISA, Blotting, DNA/RNA-hybridization and IHC applications. High Sensitivity is our motto and our commitment. Making use of complex sophisticated detection technologies associated with extra high costs is not the only way to achieve a High Sensitivity in assaying different analytes. Our reagents alone offer straightforward and practicable alternative. Products we make are essentially simple, stable, robust, easy-to-handle and easy-to-use reagents that are capable of providing exceptionally high sensitivity in immunoassays that are well known and technically also simple. These are assays of most viable and exploitable format, such as conventional ELISA and related tests. At that it does not really matter whether it is routine colorimetric ELISA with chromogens or test exploiting Enhanced ChemiLuminescence or other non-colorimetric substrate development system.

We offer three groups of Specialty Products along with miscellaneous ImmunoAssay Support (Accessory) Reagents: - Poly-HRP and PolyGOD Conjugates - Poly-HRP Diluents, Blockers and Stabilizers

- TMB Substrates

- ImmunoAssay Accessory Reagents

Together they comprise a comprehensive line of universal high performance reagents that are readily applicable in development and large-scale production of High Sensitivity ELISA test kits and related assay test systems. Poly-HRP conjugates are unique products for ultra-sensitive detection manufactured solely by SDT.

Poly-HRP detection enables low picogram femtogram sensitivity in 2-3-hour conventional colorimetric ELISA. 3D ImmunoFiltration assays yield with Poly-HRP comparable sensitivity levels in just 20-30 minutes. So if High Sensitivity is a larger claim in our mission statement, it is quite logical that Poly-HRP is our core business. We are very focused on Poly-HRP and, as a sole Poly-HRP manufacturer, keep a commitment of being reliable Poly-HRP supplier to IVD Industry and Life Science Research Community.

We produce universal

Streptavidin-Poly-HRP,
recombinant ProteinA-Poly-HRP and
recombinant ProteinA/G-Poly-HRP conjugates,
Goat anti-Mouse IgG-Poly-HRP,
Rabbit anti-Mouse IgG-Poly-HRP,
Goat anti-Rabbit IgG-Poly-HRP and
Mouse monoclonal anti-Human IgG(H,γ-chain specific)- and anti-Human IgM (H, µ-chain specific)-Poly-HRP conjugates. conjugates.

Our Streptavidin-Poly-HRP conjugates are synthesized in three forms,
Streptavidin-Poly-HRP20 (#SP20), Streptavidin-Poly-HRP40 (#SP40) and Streptavidin-Poly-HRP80 (#SP80), reflecting polymerization range and respectively size of HRP homopolymers in signal-generating moiety of detecting conjugate. Size of the Poly-HRP determines the level of detection enhancement with Poly-HRP. See Poly-HRP Application Note and Poly-HRP Users Guide for details on specific molecular design and detecting activity of different Poly-HRP conjugates.

Streptavidin-Poly-HRP satisfies highest sensitivity requirements and, along with dedicated SA-Poly-HRP Diluents/Blockers and Stabilizers, offers effective solution in rapidly designing ultra-sensitive ELISA test from scratch. This version is also advisable when only minimal amounts of specific antibody/antigen reagents are available for development. The only prerequisite here is a biotinylated anti-analyte. Biotinylation of antibody or antigen through primary aminogroups using NHX-biotin esters is perhaps the easiest conjugation technique known in immunochemistry. Besides, biotinylation can be performed well reproducibly with just a couple of milligrams and even smaller amounts of anti-analyte protein. In other words, with Streptavidin-Poly-HRP one has a fairly good chance to develop in a short time really ultrasensitive assay, e.g. two-site double antibody sandwich ELISA (as non validated model test system) having just few milligrams of purified intact capture (binding) antibody and detecting (tracer/detector) antibody-biotin conjugate along with antigen calibrator and standard ELISA equipment and accessories.

Referring to our Poly-HRP Users Guide will improve this chance. This handbook accumulates our extensive experimental experience with Poly-HRP and has been especially written to render comprehensive technical assistance to researches and developers by showing pitfalls, giving practicable advice and troubleshooting. On the common, every detection system using biotin-streptavidin interaction, although usually complying with highest sensitivity demands, still has one intrinsic disadvantage: it is two-step system by definition.

To those developers who wish to avoid using streptavidin and at the same time get a full advantage of Poly-HRP in one-step detection we offer an extensive program of Custom Poly-HRP Conjugations. This option is addressed primarily to larger OEM customers to whom starting amount of antibody or antigen is not a restraint (see Section Custom Poly-HRP Conjugation for details). PolyGOD product line is limited to Streptavidin-(mono)GOD (#SG1), Streptavidin-PolyGOD5 (#SG5) and Streptavidin-PolyGOD20 (#SG20) conjugates. Suggested application is basically with electrochemical Immunosensors, glucose and oxygen sensing, single-layer and multilayer thin-film Immunoassays, peroxide-tunnelling Immunoassay systems and IHC (double immunostaining) applications.

Our Diluents/Blockers and Stabilizers have initially been developed as complementary line of auxiliary biotin-free reagents supporting Streptavidin-Poly-HRP product line.

However, all biotin-free products can successfully be used not only with Streptavidin-Poly-HRP and not only with Poly-HRP but also with plenty of other, quite different immunoreagents and conjugates. Therefore they are useful miscellaneous products that feature excellent NSB-eliminating, boosting and stabilizing activities. Field of their application is also not restricted to only ELISA or Immunoblotting. Our Diluents/Blockers and Stabilizers have a potential to improve performance of virtually any (immuno)analytical system irrespective of specific test format through decreasing backgrounds, boosting positive signals, aggregately yielding better signal-to-noise ratios, eliminating matrix effects and storage drifts.

We have developed proprietary large-scale production process for manufacturing of highly concentrated stable 0.2/0.45-m filterable Casein Colloid Buffer which is a basic intermediate product in many our stable liquid formulations.

Universal Casein Diluent/Blocker (#UCDB), Casein Buffer Concentrate 1 (#CBC1) and both SA-Poly-HRP Stabilizers (#SA1 & #SA3-RHT) are biotin-free products. This means they are (i) made of specially selected, thoroughly proven essentially biotin-free raw material and (ii) trace endogenous, both free and bound, biotin has been completely removed in process of product preparation. Biotin-free Diluents and Stabilizers are universal non restricted products ideal for use with Streptavidin-Poly-HRP. Antibody/Antigen/Conjugate Diluent/Blocker (#AADB), Casein Buffer Concentrate 2 (#CBC2) and both Antibody/Antigen (Poly-HRP) Conjugate Stabilizers (#AA1 & #AA3-RHT) are not biotin-free. Therefore they are 30% cheaper and can be used as well as universal biotin-free products in all assay steps and with all assay reagents except for Streptavidin-Poly-HRP.

Aside from extra special functions (related to Stabilizers) they are all excellent diluents/blockers that will effectively eliminate backgrounds mediated through non-specific protein-protein binding in solution and in heterogeneous (liquid/solid phase) systems. Our TMB Substrates are much more than just newer me, too products. When we began TMB development we did well realize it might be too late for launching our own TMB product line. Indeed, there are many stable 1-component ready-to-use TMB formulations on the market and many are good enough to satisfy most customers requirements. With our given policy of endeavoring to be the very best high quality IVD reagent supplier, we would probably decide not to launch our s(HS)TMB for ELISA, if this product would not outperform the best currently available competitive products on the market. This concerns not only sensitivity but also other performance characteristics that are frequently underestimated by other manufacturers. See respective section for more detailed info and order promotional trial sample of our s(HS)TMB to make yourself convinced of its strong High Sensitivity performance advantages. p(HS)TMB is a precipitating formulation of TMB. This type of product is also frequently named membrane TMB. Although our p(HS)TMB has been developed primarily not for membrane blotting applications but for assays designed in 3D (Immuno)Filtration format it will yield as well superior sensitivity in most of 2D assays, as dot- & line-blot, deep-stick, classical blotting and ELISPOT. To facilitate developmental work with our specialty reagents we also offer a line of miscellaneous ancillary products comprising standard liquid formulations and buffers routinely used in ELISA, blotting and related immunotests.

Amid these Assay Support Reagents are  concentrated 10% BSA solutions, HAMA Blocker, Universal Blocker of heterophilic antibodies, rheumatoid factors, HAMA and complement, Cold Capture Antibody controls, concentrates of basic working buffers for ELISA and 3D-ImmunoFiltration - PBS, Coating, Wash and Substrate Buffer Concentrates. All liquid reagents and buffers are stable, reliably preserved with 5-Bromo-5-nitro-1,3-dioxane (BND), filtered through 0.2 m liquid formulations and are easy-to-use items that can reasonably speed-up (immuno)assay development process while making it comfortable to those researches and developers who are unwilling or reluctant of preparing their own buffers and liquid reagents. We additionally offer BND, very reliable anti-microbial preservative/biocide exceptionally effective against bacteria, yeast and fungi, product that features excellent performance in preserving stable liquid IVD formulations with shelf lives exceeding 12-24 months.

All our products are fully validated robust products featuring well documented real-time stability in recommended regular storage regimens within and frequently far beyond accepted in IVD industry reagent stability standards. We manufacture at our own facilities in ITC, Baesweiler, Germany
